1- Identify candidate serum and bile metabolites associated with disease severity in PSC by performing metabolimics studies.
2- To test causality of identified candidate metabolites in modulating bile duct inflammation and fibrosis using translational research models of PSC.This PhD project will employ a combination of in vitro (immortalized and primary cholangiocytes), ex vivo (patient-derived cholangioids), and in vivo (diet- and genetically induced mouse models of PSC) systems to test the hypothesis that specific metabolites contribute to bile duct inflammation and fibrosis. It is desirable that applicants have a background in biology and/or knowledge in liver (patho)physiology. Experience in cholangiocytes biology and in using in-vitro and in-vivo models (e.g. cell culture, mouse experiments, flow cytometry, qPCRS, immunostaining) is an advantage.Doctoral studies comprises four years of full-time study, and leads to a doctoral degree.
